Kaempferol, a dietary flavonoid found in fruits and vegetables, has shown remarkable potential in promoting immune tolerance and combating allergies. A recent study published in Allergy reveals how kaempferol stimulates regulatory T cell (Treg) development and suppresses inflammation by activating key immune pathways.
Regulatory T cells play a pivotal role in maintaining immune balance and preventing chronic inflammation. This study highlights its ability to activate the Aldh1a2/Raldh2 gene in intestinal dendritic cells (DCs) through the aryl hydrocarbon receptor (AhR) and transcription factors PU.1 and IRF4. These genes enable dendritic cells to convert retinal into retinoic acid, which is essential for Treg differentiation.
Researchers screened various dietary compounds and identified kaempferol as the most effective in enhancing Treg-inducing functions in DCs. Experiments demonstrated that kaempferol-treated DCs significantly increased Treg frequencies while suppressing T-cell proliferation. The study also revealed that its effects depend on its interaction with AhR, a transcription factor known to regulate immune responses. By antagonizing AhR, kaempferol alleviates its inhibitory effect on Aldh1a2 expression, enhancing Treg development.
In addition to its in vitro success, kaempferol was tested in mouse models to explore its role in managing allergic inflammation. When applied in a food allergy model, it significantly reduced allergic symptoms, such as diarrhea and sudden temperature drops, during allergen challenges. These findings suggest that kaempferol strengthens immune tolerance by suppressing inflammatory responses.
The study also uncovered how it influences PU.1 and IRF4 expression, two transcription factors critical for activating Aldh1a2. By relieving AhR-mediated repression, it enhances their activity, further boosting Treg development.
In conclusion, this research positions kaempferol as a promising dietary compound for managing immune-related conditions. Its ability to promote regulatory T cells and reduce allergic inflammation underscores its potential as a natural therapeutic agent for improving immune health.
